I can use automapping and set rules for one block. But if i want to output a pattern which require multiple input block. Do I still need regions layer?
English is not my first language so it’s kinda hard to understand.

No, you do not need a regions layer at all.
In what way does the rule “not work”? What does your input (on your working map) look like? Your rule looks for a 1x3 area of that tile, surrounded by empty cells. If that’s not the scenario in your map, then the rule won’t match and won’t output anything.
